That wording is confusing, like all of Rifts, but it's referring to normal combat turn order. You don't roll Initiative again whenever you cast a spell, that would give both enemies and allies extra actions, would be insane, and would make no sense.

Welp, no need to use the rule, because yeah, it's confusing.
What I personally get from it is that you make an initiative roll not to actually restart initiative, but just to see if you can interrupt the spell.

We'll, first of all, Instant cast spells cannot be interrupted. For obvious reasons. And like I said before, channel spells can be interrupted normally.
That interpretation makes no sense. Say I try to cast the spell Lightblade. The nearest enemy is 30 feet away from me. According to that, say I roll Initiative to cast a spell that only costs 1 action and roll low. The enemy rolls higher. Does it immediately get to teleport 30 feet and interrupt my spell? Of course not. That makes no sense.
All this talk will do is confuse Mystic more.

I think it means that other spellcasters and people with shooty guns and people next to you can try to interrupt it. I got the impression it meant for out-of-combat. Basically that if you tried to sucker-punch someone with a spell there's a chance they could go full spaghetti western and try to draw first.

The rule we're talking about is for level 1-5 spells though.

Oh, that one. I thought you meant the whole discussion. It pretty much boils down to if you're overtly casting a spell it's the same as pulling a gun on someone or something: roll initiative 'cause you just started a fight.

Pretty much what Birdy said. I posted the rules on interrupting spells normally on the last page. It's actually really simple when you finally get past the terrible wording that plagues Rifts.
